I'm not sure what you are asking. Are you concerned about how much to take with each roughing cut?

Or, if it's a question about finish depth, I think you could use a span/caliper measurement over a number of teeth, or you could use an over pin measurement.

Over pin might not be practical if the OD is larger than largest available micrometer.

Measuring depth is not a reliable way to get the correct "meshing" depth at assembly.

Span measurement using a caliper-style instrument over a predefined number of teeth is commonly used to judge proper finish cut.
